General description

Water is a critical component of all living cells. Interestingly, tissue membranes show a great degree of water permeability. Mammalian red cells, renal proximal tubules, and descending thin limb of Henle are extraordinarily permeable to water. Water crosses hydrophobic plasma membranes either by simple diffusion or through a facilitative transport mechanism mediated by special proteins "aquaporins". Aquaporins, are integral membrane proteins from a larger family of major intrinsic proteins (MIP) that form pores in the membrane of biological cells. Over the last decade, genes for several members of aquaporin family have been cloned, expressed, and their distribution studied in many tissues. The water-specific channel Aquaporin 4 is located in various tissues including the gastrointestinal tract, kidney and brain. It is an osmoreceptor which regulates body water balance and mediates water flow within the central nervous system. In CNS tissue, AQP4 localizes to astrocyte endfeet making it a useful structural marker.

Specificity

Reactivity with other species has not yet been tested. It is expected that the antibody may also react with bovine (71/75) and rabbit (64/75) due to sequence homology.
Recognizes Aquaporin 4 (AQP4, WCH4, Mercurial-Insensitive Water Channel).

Immunogen

Epitope: C-terminus
GST fusion protein with residues 249-323 of rat Aquaporin 4 (SwissProt accession number P47863).

Application

Detect Aquaporin 4 using this Anti-Aquaporin 4 Antibody, C-terminus validated for use in IH & WB.
Immunohistochemistry:
A previous lot of this antibody was used on rat brain sections.
Dilutions should be made using a carrier protein such as BSA (1-3%).

Western blotting:
1:500-1:1,000 using ECL on rat brain membranes.

Optimal working dilutions must be determined by the end user

ImmunoAffinity Purified: Amber vial of Rb X Aquaporin 4, C-Terminus (part # AB3594-50ULa)
Purified rabbit polyclonal, lyophilized from buffer containing PBS, pH 7.4, containing 1% BSA, and 0.05% sodium azide. Reconstitute with 200 µL of sterile distilled water. Centrifuge antibody preparation before use (10,000 x g for 5 min).

Storage and Stability

Stable for up to 6 months at -20ºC from date of receipt. After reconstitution stable at -20ºC in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Analysis Note

Control
Clear vial of RbX Aquaporin 4, CT Control Antigen(AB3594-50ULb) included free of charge with the antibody is 120 µg of control antigen (lyophilized powder). Reconstitute with 100 µL of PBS. For positive control, in Western blot use 20 ng of protein per Minigel lane. For negative control, preincubate 3 µg of purified peptide with 1 µg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
