Quality
Routinely evaluated by phosphorylation of 0.8 mg/mL myelin basic protein. Purity was assessed by SDS-PAGE and Coomassie blue staining using 3 μg of TAO1.
Unit Definition
Specific Activity: where one unit of TAO1 activity is defined as 1 nmol phosphate incorporated into 0.8 mg/mL myelin basic protein per minute at 30°C with a final ATP concentration of 100 μM.
Physical form
10 μg of enzyme in 100 μL of 50 mM Tris/HCl pH7.5, 300 mM NaCl, 0.1 mM EGTA, 0.03% Brij 35, 270 mM sucrose, 1 mM benzamidine, 0.2 mM PMSF, 0.1% 2-mercaptoethanol. Frozen solution.
Ni2+/NTA-agarose
Storage and Stability
Stable for 6 months at -70°C from date of shipment. For maximum recovery of product, centrifuge original vial prior to removing the cap. Rapidly thaw the vial under cold water and immediately place on ice. Aliquot unused material into pre-chilled microcentrifuge tubes and immediately snap-freeze the vials in liquid nitrogen prior to re-storage at -70°C.

Several components of the budding yeast pheromone-response pathway are conserved in mammalian mitogen-activated protein (MAP) kinase pathways. Thus, we used degenerate oligonucleotides derived from the sequence of the Saccharomyces cerevisiae protein kinase Ste20p to amplify related sequences from the rat.
