Specificity

Clone 1D18 is a ZooMAb® rabbit recombinant monoclonal antibody that detects Insulin-like growth factor 1 receptor. It targets an epitope within 21 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal region of human IGF-1R β chain.

Application

Quality Control Testing

Evaluated by Western Blotting in NIH3T3 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IGF-1R β in NIH3T3 cell lysate.

Tested applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected IGF-1R β in A549 and C6 cell lysates.

Flow Cytometry Analysis: 0.1 μg from a representative lot detected IGF-1R β in one million A549 and NIH3T3 cells.

Affinity Binding Assay: A representative lot of this antibody bound IGF-1R beta peptide with a KD of 1.1 x 10-7 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected IGF-1R β in human breast cancer and human colon cancer tissue sections.

Target description

Insulin-like growth factor 1 receptor (UniProt: P08069; also known as EC:2.7.10.1, Insulin-like growth factor I receptor, IGF-I receptor, IGF1-R, CD221) is encoded by the IGF1R gene (Gene ID: 3480) in human. IGF-1R is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-30), which is subsequently cleaved off to generate the mature form that is cleaved into an -chain (aa 31-736) and a β-chain (aa 741-1367). IGF-1R is a tetramer of two - and two β-chains linked by disulfide bonds. The -chains contribute to the formation of the ligand-binding domain, while the β-chain carries the kinase domain. The β-chain has an extracellular domain (aa 741-935), a transmembrane domain (aa 936-959), and a cytoplasmic domain (960-1367). IGF-1R binds to IGF-1 with high affinity and with IGF-2 and insulin with a lower affinity. In response to ligand binding, it is autophosphorylated on Tyr 1165, Tyr 1161, and Tyr 1166 in a sequential manner. Its autophosphorylation occurs in a trans manner where one subunit of the dimeric receptor phosphorylates tyrosine residues on the other subunit. While every single phosphorylation increases kinase activity, all three tyrosine residues in the kinase activation loop have to be phosphorylated for optimal activity. The activated IGF-1R is involved in cell growth and survival control, and it is also crucial for tumor transformation and survival of malignant cell. This ZooMAbZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
