Chromenone 1 is a bone morphogenetic protein (BMP) signaling potentiator that exerts its effect via negative TGFβ feedback loops, independent of direct kinase inhibition. Chromenone 1 prevents cardiogenesis induction by 0.5 μM BMP inhibitor DMH1 (IC50 = 200 nM; mESC-Myh6-GFP) and efficiently stimulates BMP-dependent osteogenesis from C2C12 myoblasts (ALP activity induction EC50 = 340 nM; E100% and Emax = induction by 7.5 and 50 ng/mL BMP-4, respectively).
