Cell Line Description

The drug-resistant cell line EMT6/MTXR was derived from the mouse mammary tumour cell line EMT6/P (Sigma Catalogue number. 96042344) by growth in increasing concentrations of methotrexate. EMT6/MTXR did not hyperexpress P-glycoprotein and did not show cross-resistance to other drugs of the multi-drug resistance (MDR) phenotype. It is recommended to culture the cells without drug on resuscitation until first passage or growth has been fully established.

Culture Medium

MEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 1μg/ml methotrexate + 15% Foetal Bovine Serum (FBS).. Resuscitate in drug-free medium; add drug once growth is fully established

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 using 0.25% trypsin or trypsin/EDTA;5% CO2; 37ºC
