General description

Fluorescent transfection reagent has the cationic lipid 1,2-dioleoyloxypropyl)-N,N,N-trimethylammonium chloride (DOTAP) and the neutral lipid DOPE (1,2-dioleoyl-sn-glycero-3-phosphoethanolamine in the ratio 1:1 w/w along with 0.1 wt% N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)dioleoyl (NBD) DOPE. NBD-DOPE is a fluorescence probe with NBD attached to headgroup of DOPE.

Biochem/physiol Actions

The DOTAP:DOPE with 0.1% NBD-DOPE liposome (LipNBD) reconstituted with transferrin called the transferrin liposome-nitrobenzoxadiazole (Tf-LipNBD) is useful as a probe. It provides improved image contrast in in vitro magnetic resonance imaging (MRI) of tumor cells. Fluorescent transfection reagent aids in understanding fibroplex membrane structure and fibroin interaction with cationic lipids. N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)dioleoyl (NBD) is used in model membranes studies and is useful in visualizing membrane domains.
