Spezifität

Clone 3L23 is a ZooMAb® recombinant rabbit monoclonal antibody that detects Nibrin/NBS1. It targets an epitope within the C-terminal half.

Immunogen

His tagged, recombinant fragment corresponding to 335 amino acids from the C-terminal half of human Nibrin/NBS1.

Anwendung

Quality Control Testing

Evaluated by Western Blotting in nuclear extract from HeLa cells.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected Nibrin/NBS1 in nuclear extract from HeLa cells.

Tested Applications

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Nibrin/NBS1 in HeLa cells.

Affinity Binding Assay: A representative lot of this antibody bound Nibrin/NBS1 protein with at least one thousand-fold (1,000X) higher than with non-specific GST control.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Nibrin/NBS1 in human testis tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Zielbeschreibung

Nibrin (UniProt: O60934; also known as Cell cycle regulatory protein p95, Nijmegen breakage syndrome protein 1, hNbs1) is encoded by the NBN (also known as NBS, NBS1, P95) gene (Gene ID: 4683) in human. Nibrin consists of multiple functional domains including forkhead-associated (FHA) and BRCA1 C-terminus (BRCT) domains. The protein typically forms a complex called MRN, along with MRE11 and RAD50. Nibrin is primarily involved in the cellular response to DNA damage, particularly in the repair of double-strand breaks through homologous recombination and non-homologous end joining pathways. It acts as a signal transducer by recognizing sites of damage and recruiting other proteins to the site, facilitating DNA repair. Nibrin also plays a role in maintaining telomere integrity and in the activation of cell cycle checkpoints in response to DNA damage. It is primarily localized in the nucleus. Upon DNA damage, it is recruited to the sites of double-strand breaks where it co-localizes with other DNA repair machinery components. Mutations in the NBN gene are directly linked to Nijmegen Breakage Syndrome (NBS), a rare autosomal recessive disorder characterized by microcephaly, immunodeficiency, radiosensitivity, and a predisposition to cancer. Individuals with NBS typically have mutations that lead to truncated forms of nibrin, impairing its ability to effectively participate in DNA repair processes. This deficiency in DNA repair mechanisms significantly contributes to the disease pathogenesis, including developmental abnormalities and cancer predisposition. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physikalische Form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Rekonstituierung

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Lagerung und Haltbarkeit

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
