Allgemeine Beschreibung

The gene CARD14 (caspase recruitment domain family member 14) encodes a member of the MAGUK (membrane-associated guanylate kinase) family of proteins. These proteins are scaffold proteins that participate in the assembly of multiprotein complexes at certain sites on the plasma membrane. The encoded protein contains an N-terminal CARD domain, a central coiled-coil domain, and a tripartite domain comprising of a PDZ (PSD-95, Dlg1, ZO-1) domain, an Src (Sarcoma) homology 3 domain, and a GUK (Guanylate kinase) domain at the C-terminus. The gene is mapped to human chromosome 17q25. It is mainly expressed in the basal and suprabasal layers of healthy skin epidermis.

The gene CARD14 (caspase recruitment domain family member 14) encodes a nuclear factor that contains a CARD domain involved in binding to the CARD domain of BCL10 (B cell leukemia 10), a protein involved in the activation of NF-κB through the IKK complex, and positive regulation of apoptosis. Mutations in this gene are associated with familial pityriasis rubra pilaris, a papulosquamous disorder phenotypically related to psoriasis. It also activates p38 and JNK MAP (c-Jun N-terminal kinase mitogen-activated protein) kinase pathways via association with paracaspase-1 (PCASP-1), also referred to as MALT1, which can serve as a therapeutic target in psoriasis.
