Allgemeine Beschreibung

DNA repair protein XRCC1 (UniProt: P18887; also known as X-ray repair cross-complementing protein 1) is encoded by the XRCC1 gene (Gene ID: 7515) in human. XRCC1 is a scaffold protein that is involved in DNA single-strand break repairs by mediating the assembly of DNA break repair protein complexes. Upon detection of DNA damage, it moves from the nucleoli to the global nuclear chromatin. It specifically interacts with nicked and gapped DNA and acts as a nick sensor capable of interacting with a variety of other proteins involved in repair, including DNA polymerases and DNA ligases. It contains three distinct domains: the N-terminal domain, and two independent BRCT domains (aa 315-403 and 539-629), connected by two intervening unstructured linkers that serve as critical interacting platforms for various repair factors. The first BRCT domain is reported to interact with PARP1, APE1, MPG, NTH1 and NEIL1 and the second BRCT domain interacts with DNA ligase III. It is shown to negatively regulate ADP-ribosyltransferase activity of PARP1 during base-excision repair in order to prevent excessive PARP1 activity. (Ref.: Mok, MCY., et al. (2019). Sci. Anwendung

Quality Control Testing

Evaluated by Western Blotting in HeLa cell nuclear extract.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected XRCC1 in HeLa cell nuclear extract.

Tested Applications

Western Blotting Analysis: A representative lot detected XRCC1 in Western Blotting application (Caldecott, K.W., et al. (1995). Nucleic Acids Res. 23(23):4836-43).

Immunoprecipitation Analysis: A representative lot immunoprecipitated XRCC1 in Immunoprecipitation application (Caldecott, K.W., et al. (1995). Nucleic Acids Res. 23(23):4836-43).

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.
