Beschreibung

General description

Ein zellpermeables Hydroxyethyl-Dipeptid-Isoster, das als potenter und hochspezifischer Inhibitor der γ;-Sekretase wirkt (Aβ;gesamt IC₅₀ = 17 nM, Aβ;₄₀ IC₅₀ = 48 nM und Aβ;₄₂ IC₅₀ = 67 nM in SH-SY5Y-Zellen mit spβ;A4CTF-Überexpression). Bindet an Presenilin und blockiert die Produktion der intrazellulären Notch-Domäne. Laut Berichten wird auch die Bildung von ε;CTF blockiert, was eine Akkumulation von α;- und β;CTF bewirkt. Dient als Übergangsstatus-Analogsimulation des katalytischen Zentrums der Aspartylprotease, bietet jedoch eine 100-fach größere Selektivität für γ;-Sekretase als für Cathepsin D.
