Specificity

Clone 1E10 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects IRF-4. It targets an epitope within 21 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 21 amino acids from the N-terminal half of human IRF-4.

Application

Quality Control Testing

Evaluated by Western Blotting in HuT 78 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected IRF-4 in HuT 78 cell lysate.

Tested Applications

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected IRF-4 in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound IRF-4 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected IRF-4 in Ramos cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Interferon regulatory factor 4 (UniProt: Q15306; also known as IRF-4, Lymphocyte-specific interferon regulatory factor (LSIRF), Multiple myeloma oncogene 1, NF-EM5) is encoded by the IRF4 (also known as MUM1) gene (Gene ID: 3662) in human. IFN regulatory factor (IRF)-4 is a lymphoid/myeloid-restricted member of the IRF transcription factor family that plays an essential role in the homeostasis and function of mature lymphocytes. It has a N-terminal DNA-binding domain (DBD) characterized by a helix-turn-helix motif that enables binding to specific DNA sequences known as interferon-stimulated response elements (ISREs). IRF4 is predominantly localized in the nucleus, where it binds to DNA and regulates gene transcription. IRF4 expression is primarily observed in immune cells, including B cells, T cells, macrophages, and dendritic cells and the expression is tightly regulated in resting primary T cells and is transiently induced at the mRNA and protein levels after activation by Ag-mimetic stimuli such as TCR cross-linking or treatment with phorbol ester and calcium ionophore (PMA/ionomycin). IRF4 modulates the expression of various cytokines and cytokine receptors, influencing immune cell differentiation and function. Mainly, it is essential for the differentiation of B cells into plasma cells and aslo, influences the development of T-helper cell subsets, including Th1, Th2, Th17, and regulatory T cells (Tregs). In deficiency of IRF4, B cells shows impaired expression of activation-induced deaminase and lacks class-switch recombination. Aberrant expression or mutations of IRF4 are associated with various hematological malignancies, including multiple myeloma, diffuse large B-cell lymphoma, and chronic lymphocytic leukemia. Dysregulation of IRF4 has been linked to autoimmune conditions such as systemic lupus erythematosus and rheumatoid arthritis due to its role in immune cell differentiation and function. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
