General description

Specificity

Clone 1B13 is a ZooMAb® rabbit recombinant monoclonal antibody that detects ApoER2. It targets an epitope within 18 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 18 amino acids from the C-terminal half of human ApoER2.

Application

Quality Control Testing

Evaluated by Western Blotting in U-87 MG cell lysate.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected ApoER2 in U-87 MG cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected ApoER2/LRP-8 in HT-29 cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected ApoER2/LRP-8 in human testis tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound ApoER2/LRP-8 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected ApoER2/LRP-8 in HT-29 cells.

Target description

Low-density lipoprotein receptor-related protein 8 (UniProt: Q14114; also known as LRP-8, Apolipoprotein E receptor 2) is encoded by the LRP8 (also known as APOER2) gene (Gene ID: 7804) in human. ApoER2 is a member of the LDL receptor family, which is characterized by distinct structural motifs, including an N-terminal ligand-binding domain with multiple cysteine-rich repeats, EGF precursor homology domains containing YWTD β-propeller, an O-linked sugar domain near the membrane-spanning segment, and a cytoplasmic domain containing the NPXY motif. ApoER2 is involved in mediating cellular uptake of lipoprotein particles and is integral to signal transduction pathways influencing neuronal migration, synaptic plasticity, and cell signaling. It interacts with several ligands, including Apolipoprotein E, Reelin, and alpha-2-macroglobulin, among others. LRP8 is ubiquitously expressed but shows a higher expression level in certain tissues like the brain, where it plays a significant role in neurodevelopment and synaptic functions. Its expression can be regulated at both the transcriptional and post-transcriptional levels, influenced by cellular conditions and developmental stages. ApoER2 has been implicated in several diseases, including Alzheimer′s disease, atherosclerosis, and cancer. Its dysregulation has been associated with altered lipid metabolism, impaired synaptic function, and abnormal cellular proliferation and migration.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
