Specificity

Clone DM1A is a ZooMAb® mouse recombinant monoclonal antibody that detects a-Tubulin in multiple species. It targets an epitope within 24 amino acids from the C-terminal region.

Application

Quality Control Testing

Evaluated by Western Blotting in MCF-7 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected a-Tubulin in MCF-7 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected a-Tubulin in NIH3T3 and PC12 cell lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected a-Tubulin in human kidney and human testis tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected a-Tubulin in HeLa, A431, HepG2, and NIH 3T3 cells.

Anti-a-Tubulin, clone DM1A ZooMAb®, Cat. No. ZMS1039, is a recombinant Mouse monoclonal antibody that specifically targets a-Tubulin and is tested for use in Immunocytochemistry, Immunohistochemistry (Paraffin), and Western Blotting.

Target description

Tubulin alpha-1A chain (UniProt: Q71U36; also known as Alpha-tubulin 3, Tubulin B-alpha-1, Tubulin alpha-3 chain) is encoded by the TUBA1A (also known as TUBA3) gene (Gene ID: 7846) in human. Tubulin is the major constituent of microtubules (MTs). MTs are cytoskeletal filaments composed of  a /β-tubulin heterodimers. Tubulin dimers constantly polymerize and depolymerize, and MTs can undergo rapid cycles of assembly and disassembly. In the presence of Mg2+ and GTP, a and β tubulins join in an end-to-end manner to form protofilaments with alternating a and β subunits. Tubulin binds two moles of GTP, one at an exchangeable site on the ß-chain and one at a non-exchangeable site on the a-chain. GTP bound to a-tubulin is hydrolyzed to GDP during or immediately after polymerization. This weakens the binding affinity of tubulin for adjacent molecules and favors depolymerization that contributes to the dynamic behavior of MTs. MTs also undergo treadmilling, in which tubulin molecules bound to GDP are continually lost from the minus end and are replaced by the addition of GTP-bound tubulin molecules to the plus end of the same MT. Various post-translational modifications, including acetylation, polyglutamylation, polyglycylation, detyrosination, phosphorylation, and palmitoylation, can regulate the polymerization properties of tubulins and/or their interactions with microtubule associated proteins and motor proteins. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
