Bacillus cereus E33L ... BCZK3513(3026815)

General description

Phosphatidylinositol-specific phospholipase C (PI-PLC) from Bacillus cereus is a 298 residual protein. It belongs to the class of ubiquitous enzymes. PI-PLC is characterized with a single globular domain and it folds into eight (βα) barrel sheets, out of which only six βα units are found in barrel, lacking helices between strands IV and V and V and VI.

Application

Phospholipase C, Phosphatidylinositol-specific from Bacillus cereus has been used to cleave GPI-anchored protein from green fluorescent protein- glycosylphosphatidylinositol (GFP-GPI) from the cell membrane.[1] It has also been used in invasion assay to test the involvement of PLAP in T. cruzi invasion to human epithelial type 2 (HEp-2) cells.

Biochem/physiol Actions

Used for the release of GPI anchored proteins from the membrane.

Physical form

Contains phosphate buffer salts, EDTA and stabilizer

Analysis Note

Acetylcholinesterase is measured according to Ellman, et al.

Other Notes

One unit will liberate one unit of acetylcholinesterase per minute from a membrane-bound crude preparation at pH 7.4 at 30 °C (10 minute incubation).

Questions

  1. Which buffer should I use to reconstitute the enzyme in aqueous form and store at 4C or -20C?

    1 answer
    1. The enzyme can be solubilized in 10 mM Tris-HCl, pH 7.4, containing 144 mM NaCl and 0.05% BSA. Store this product at –20 °C. The lyophilized product, when stored properly, retains activity for at least 2 years. A preparation of this enzyme in 60% (v/v) glycerol containing 10 mM Tris-HCl, pH 8.0, and 10 mM EDTA retains activity at 2–8 °C for at least 3 years.
