Application

Neocarzinostatin from Streptomyces carzinostaticus has been used:
  • as a DNA damaging agent to treat human fibroblast GM0637 cells
  • as a radiomimetic antibiotic to induce DNA damage[1]
  • in combination with Vγ2Vδ2 T cells to determine the cytotoxicity in ovarian cancer cells

Biochem/physiol Actions

Neocarzinostatin is a protein-small molecule complex composed of an enediyne chromophore tightly bound to a 113 amino acid single chain protein. The complex possesses antiproliferative and antitumor activity. The chromophore is the active compound, which is responsible for DNA cleavage; while the apoprotein stabilizes and regulates the availability of the labile chromophore. NCS chromophore is bound non-covalently in a cleft of the binding protein and is dissociable. Upon addition of a thiol, the chromophore forms a highly reactive biradical species that can induce sequence-specific single and double strand breaks in DNA. Neocarzinostatin inhibits DNA synthesis and possesses antitumor activity in various human and animal tumors. NCS inhibits cellular proliferation by inducing G2 cell cycle arrest and apoptosis in both human papillomavirus (HPV) positive and negative cell lines.
Neocarzinostatin is a radiomimetic antibiotic substance.[1]

  1. By mistake, I stored only for 24hs at -20. I put it again at 2-8C. It's should still be active?

    1. Unfortunately, the product storage Instructions published in the product datasheet specifically note 'do not freeze'. The freeze/thaw process is detrimental to the quality of this material. The decline in activity upon thawing cannot be predicted. Please see the link below to review the product datasheet:

  2. I have Neocarzinostatin (sigma,N9162). I will prepare different concentrations but i am not sure how to calculate it. The data sheet says that protein concentration is 0.5 mg/ml. The product is 100 ug and the molecular weight of neo is 659.64 g/mol

    1. The protein content specification for this product is 0.50 - 0.70 mg/ml and will vary from lot to lot. The lot specific value may be found in the Certificate of Analysis.       See below to review an example scenario:
      If the concentration of the 100µg package is 0.50 mg/ml (500µg/mL) protein concentration the volume supplied is 200µl.

      At a molecular weight of 659.64 g/mol the Molarity of this solution will be 757.9892 µM.
      100µg = X micromolar X 200 µl X 659.64
      The end-user can make desired concentration from this stock.

      To prepare a specific Molar concentration see the example below:
      10 µg/ml solution from this stock, it will be 15.15 µM
      to obtain 10 µg of the product, the end-user will need
      100 : 10 : ? : 200 = 10X200/100 = 20 µl.

      To make 10 µg/ml stock from the product supplied, the end-user will need to take 20 µl of the stock which is 757.9892 µM and make up the volume to 1000µl. This will give a solution that is 10mg/ml, with 15.15 µM concentration.
