General description

Insulin is a polypeptide hormone mainly secreted from pancreas when glucose level changes in the plasma. It is an anabolic hormone and enhances the generation of storage substances, for instance glycogen, fat and protein. It also induces glucose uptake by peripheral tissues, thereby controlling glucose levels in the plasma. Its main sites of action are liver, adipose tissue and muscle.

Application

Insulin-FITC labeled human has been used in primary endothelial cells from human adipose tissue microvasculature (HAMEC) to study insulin transcytosis.[1]

  1. What is the solubility of this FITC insulin and what solvent should be used?

    1. The solubility of this product has not been tested in-house. This product is Insulin-FITC supplied as a lyophilized powder containing sodium phosphate buffer salts and is expected to be soluble in water. Based on quality control testing, a similar product is soluble in water at a concentration of 2.5 mg/ml. Insulin is generally dissolved in mildly acidic solutions.

    1. Regarding the location of the FITC label and any effects on activity, there is no specific description available on where the FITC label is positioned on FITC-insulin. However, in practice, there are only three potential sites where the FITC can derivatize the insulin. These locations include both of the N-terminal residues, off the free amine, and the Lysine at the 3rd position near the C-terminus of the B-chain.
