Hyaluronidase degrades hyaluronan and has been found to be inappropriately regulated during cancer progression.
These enzymes randomly cleave β-N-acetylhexosamine-[1→4] glycosidic bonds in hyaluronic acid, chondroitin, and chondroitin sulfates.

Other Notes

One unit will degrade 0.75 μg of the polysaccharide hyaluronic acid per minute at pH 5.35 at 37 ºC (as measured by turbidimetric absorbance (λ: 600nm) when complexed with BSA after 45 minutes).

  1. Does 1mg/ml represent the minimum solution to have a 1mg of hyaluronidase powder completely dissolved? Can the entire containing-powder vial tolerate repeated defrost-thaw cycles?

    1. It is recommended that fresh solutions be prepared before use. Stability testing of frozen solutions of this product has not been performed in our laboratories. One published reference cites storage of 1.0 mg/ml stock solutions of hyaluronidase in PBS at –20°C.   2. What should we set the specific activity of this product to? The range is 750-3,000 U/mg. We need 100 U/mL, how should we calculate it?

    1. The activity of this material is lot specific and reported in the Certificate of Analysis. The product may be reconstituted to the required concentration, based on the lot specific activity, using 20 mM sodium phosphate buffer, pH 7.0, with 77 mM NaCl and 0.1 mg/mL BSA. At the minimum activity, a 100 U/mL solution would be reconstituted with 7.5 mL of the recommended buffer, while at the maximum activity the volume would increase to 30 mL.   4. Can stock solutions of Product H3884, Hyaluronidase from bovine testes, be stored frozen after reconstitution?

    1. Although Sigma-Aldrich has not tested the stability of stock solutions of Product H3884, Hyaluronidase from bovine testes, several publications cite storage of solutions of H3884 at -20 °C:1.  Campbell, Keith H.S, "Transgenic Sheep form Cultured Cells", from Transgenesis Techniques: Principles and Protocols, Methods Mol. Biol., 180, 289-301 (2002).  A 1500 IU/mL stock solution was stored in 20 mL aliquots at -20 °C.2.  Marschall, S. and de Angelis, M.H., "In Vitro Fertilization/Cryopreservation", from Transgenic Mouse: Methods and Protocols, Methods in Molecular Biology, 209, pp. 35-50. Humana Press (2003).  1 mg/mL stock solutions in M2 medium or water, sterile-filtered (0.22 mm filter) can be stored in aliquots at -20 °C.

  5. How does the storage temperature relate to shipping conditions?

    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.
