General description

Heparin lithium is a highly sulfated polysaccharide that belongs to the family of glycosaminoglycans with remarkable significance in the areas of cell culture, cancer biology, biochemical, and drug discovery research. Due to its interactions with proteins and cells, it is used to study molecular and cellular processes, tumor modeling, investigate binding interactions, and as a cell culture supplement, to maintain cell viability.

Application

Heparin lithium was used:
  • To validate photometric measurements in cell surface charge determination
  • As an anti-coagulant for laboratory studies
  • To study hypothermal stress on lactate metabolism[1]
  • To study the effects of anticoagulants in direct membrane feeding assays
  • For 2D and 3D vitro cell culture in glioblastoma cell models[2]
  • To prevent clot formation and tissue damage during mechanical probing of cardiac pacemaker cell embryonic patterning, a study that contributes to the broader area of cardiac tissue mechanics research and developmental biology[3]

Features and Benefits

  • Heparin with specific activity of ≥150 USP units/mg
  • Versatile and adaptable for a wide variety of laboratory and research applications

Questions

1–7 of 7 Questions
  1. Could you provide the procedure for making a 20 IU/mL solution in saline using the H0878-100KU? e.g.) 1) Add 5 Liter of saline in H0878-100KU 1 bottle (volume/weight, %) 2) Vortex thoroughly 3) Filtration (0.2 um pore size) 4) Keep it at 2-4'C

    1 answer
    1. In order to prepare the solution at 20 IU/mL, the activity would be required. The activity of this product is lot specific and reported on the Certificate of Analysis. The minimum activity for any lot would be 150 units/mg. See the link below to review a sample or access the Certificate of a specific lot:
      https://www.sigmaaldrich.com/product/sigma/h0878#product-documentation

This material is soluble at 40 mg/mL in water. The same rate would be expected in saline, but rates may vary slightly. Stirring is recommended, however vigorous vortexing is not typically required.
Please see the link below to review the product data sheet which includes information on solubility and solution stability:

  3. There is no expiration date on our heparin lithium salt. The lot # is 76H1236. Is this product usable?

    1 answer
    1. This product is not assigned an expiration date. It is assigned a recommended retest date of roughly 2 years after the quality release date. The lot number given (76H1236) was released in 1996. The contents may demonstrate some activity if the bottle is unopened, however it is unlikely that it would meet specifications. A recommended retest date is the period of time during which the product is expected to remain within established stability specifications, provided that it has been stored under defined conditions. After the Retest Date, product samples should be examined to ensure that the product is still in compliance with the established specifications. For more information, you may access the "Product Dating Information" document under "ADDITIONAL USEFUL DOCUMENTS ABOUT OUR PRODUCTS" at the bottom of the Quality Services page with this link: https://www.sigmaaldrich.com/US/en/life-science/quality-and-regulatory-management/quality-services.
