General description

Glucose provides the primary energy source for cell metabolism. It is a main source of energy for living organisms. Glucose occurs naturally in the free state in fruits and other parts of plants. It is combined into glucosides, disaccharides, oligosaccharides, the polysaccharides (cellulose and starch), and glycogen.

Application

D-(+)-Glucose solution has been used:

  • to determine extracellular acidification rate (ECAR) or oxygen consumption rate (OCR) in human CB CD34+cells by Seahorse extracellular flux assay[1]
  • as a negative control in human umbilical vein endothelial cell (HUVEC) culture[2]
  • as a supplement for culturing colonic biopsies[3]

Biochem/physiol Actions

Glucose is an aldosic monosaccharide and acts as an important component of glycolysis and downstream pathways for aerobic and anaerobic respiration. It provides energy for growth and reproduction. Glucose is a byproduct of photosynthesis that is condensed to starch in plants and cyanobacteria.[4]

Other Notes

This product has been tested with cell lines to verify the product is not cytotoxic. The growth promoting capacity of this cell culture tested glucose solution was assessed at 45 mL/L in medium using appropriate cell lines.
This solution has been sterilized by filtration.

Questions

1–5 of 5 Questions
  1. Hello what is the recommended concentrations of D-glucose solution 100g/l to used for primary cell culture of brain ?

    1 answer
    1. There are a number of cell types present in brain tissue. Glucose requirements may vary by cell type. This product is not tested for use in primary cell culture. However, the typical glucose concentration in most classical media is 1 g/L or 5 g/L. In brain slice or hippocampal slice studies, the glucose concentration may vary broadly. It is advisable to search to the literature for the appropriate concentration of D-glucose to use for primary brain cell culture. Please see the links below to review examples of these publications:

  2. What is the Molar concentration of Product G8769, D-(+)-Glucose solution?

    1 answer
    1. The molar concentration of Glucose solutions G8644 and G8769, may only be estimated. The glucose molecular weight is 180.16 g/mol, however the concentration is lot specific.

      Product G8644 has a concentration specification ranging from 90 - 110 g/L or 0.50 - 0.61 M.

Please see the link below to access a sample or lot-specific Certificate of Analysis:

  3. How does the storage temperature relate to shipping conditions?

    1 answer
    1. The storage conditions that a Sigma-Aldrich catalog and label recommend for products are deliberately conservative. For many products, long-term storage at low temperatures will increase the time during which they are expected to remain in specification and therefore are labeled accordingly. Where short-term storage, shipping time frame, or exposure to conditions other than those recommended for long-term storage will not affect product quality, Sigma-Aldrich will ship at ambient temperature. The products sensitive to short-term exposure to conditions other than their recommended long-term storage are shipped on wet or dry ice. Ambient temperature shipping helps to control shipping costs for our customers. At any time, our customers can request wet- or dry-ice shipment, but the special handling is at customer expense if our product history indicates that the product is stable for regular shipment.
