Application

Silver Enhancer Kit has been used to determine the presence of gold nanorods in various tissues and to prepare silver enhanced gold colloids for electron microscopy.

Biochem/physiol Actions

Solution A and Solution B are mixed 1:1 immediately before use and applied to the slide holding the gold-labeled section. After 5-10 minutes, the slide is rinsed with distilled water, fixed for 2-3 minutes in a sodium thiosulfate solution, and rinsed again. The section is now ready for counterstaining and mounting as required.

Features and Benefits

Colloidal gold labels are normally visible only at the electron microscope level. Silver Enhancer enlarges the gold colloid label by precipitation of metallic silver to give a high contrast signal visible under a light microscope. A darkroom is not necessary for this procedure, as the silver enhancer working solution is light-stable for 20-30 minutes under ordinary laboratory lighting.

Packaging

The Silver Enhancer kit consists of Solution A (silver salt), Solution B (initiator), Sodium Thiosulfate (fixer), and a Technical Bulletin describing their use.

  1. What is the fixer Sodium Thiosulfate in Ag enhancer kit SE100 used for?

    1. Sodium thiosulfate is used to remove any unreacted silver to prevent increased background staining, per the Instructions For Use. These instructions also specify the suitability of well-prepared tissue sections cut 5–6 microns and fixed in neutral buffered formalin for iron staining, as well as the preparation method for blood or bone marrow films.

  2. For the Silver Enhancer Kit (Cat # SE100), what is the capping agent on the resulting silver nanoparticle that is formed after the silver enhancement reaction?

    1. Am not sure what is meant by the capping reagent. The Silver Enhancer kit shares similarities with the Grocott Methanamine Silver Nitrate (GMS) procedure for fungus, basement membranes or urates. The GMS and SE100 kit are both silver stain methods. The SE100 has been formulated to stain gold nanoparticles instead of fungi, basement membranes, or urates. The SE100 has 3 reagents. Reagent 1 is a silver nitrate solution. Solution 2 is a Tungstosilicic Acid Hydrate solution. This compound likely replaces the methenamine reagent typically used in the GMS procedure. Gold Chloride is considered a toning solution in the GMS procedure. There is not an equivalent reagent in the SE100 procedure. The third reagent in the SE100 kit is a sodium thiosulfate solution which removes any unreduced silver. It is likely that the Tungstosilicic Acid Hydrate is what you would refer to as a capping agent.
