General description

Mannitol Lysine Crystal Violet Brilliant Green Agar (MLCB Agar) is formulated for the selective isolation of Salmonella species, excluding Salmonella typhi and Salmonella paratyphi A, from fecal and food products. The medium contains peptic digest of animal tissue, beef extract, and yeast extract, which provides essential nutrients to support the growth of Salmonella. Mannitol serves as the fermentable carbohydrate, enabling the detection of fermentation activity, while L-lysine acts as the amino acid that facilitates lysine decarboxylation, a key metabolic process in Salmonella identification. This selective medium is designed to enhance the isolation and differentiation of Salmonella species in complex sample matrices.

Application

Mannitol Lysine Crystal Violet Brilliant Green Agar is used for isolating Salmonella species, excluding Salmonella typhi and Salmonella paratyphi A, from fecal samples and food products. It is widely applied in clinical testing for disease diagnosis, the food and beverage industry for ensuring microbiological safety, veterinary testing for disease control in animals, and microbiology research for studying Salmonella behavior in various environments.

Preparation Note

Suspend 49.02 grams in 1000 ml distilled water. Heat to boiling to dissolve the medium completely. DO NOT AUTOCLAVE. Cool to 45-50°C. Mix well and pour into sterile Petri plates.

Other Notes

Salmonellae grow as large purple-black colonies with black centers due to hydrogen sulfide production, facilitated by sodium thiosulfate and ferric ammonium citrate. Mannitol fermentation produces acid, stimulating lysine decarboxylation, which elevates the pH and promotes blackening. Atypical Salmonella strains producing little or no hydrogen sulfide may appear as mauve-grey colonies with a central black "bull′s eye." Mannitol Lysine Medium can be directly inoculated or used after enrichment in Modified Semisolid RV Medium Base. For better detection of atypical strains, Brilliant Green Agar (Modified) or Bismuth Sulfite Agar should be used alongside Mannitol Lysine Medium.
