Specificity

Clone 1B19 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects CD8 alpha. It targets an epitope with in 143 amino acids from the extracellular domain.

Immunogen

His-tagged recombinant fragment corresponding to 143 amino acids from the extracellular domain of human CD8 alpha.

Application

Anti-CD8a, clone 1B19 ZooMAb, Cat. No. ZRB1010, is a recombinant Rabbit Monoclonal Antibody that specifically targets T-cell surface glycoprotein CD8 alpha chain and has been tested for use in Immunocytochemistry, Immunofluorescence, Immunohistochemistry (Paraffin), and Western Blotting.
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected CD8a in THP-1 cell lysate.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected CD8a in human tonsil tissue sections.

Immunocytochemistry Analysis: A 1:50 dilution from a representative lot detected CD8a in THP-1 cell line.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected CD8a in human tonsil tissue sections.

Target description

T-cell surface glycoprotein CD8 alpha chain (UniProt: P01732; also known as T-lymphocyte differentiation antigen T8/Leu-2, CD8a) is encoded by the CD8A (also known as MAL) gene (Gene ID: 925) in human. CD8a is a single-pass type I integral membrane protein with one Ig-like V-type domain (aa 22-135) that plays an essential role in the immune response and serves multiple functions in responses against both external and internal offenses. CD8a is produced with a 21-amino acid signal peptide that is subsequently cleaved to produce the mature form. CD8 is generally present as a heterodimer of alpha and beta chains linked by two disulfide bonds, however, it can also form homodimers. The CD8 alpha/alpha homodimer is expressed on a subset of gamma/delta TCR-bearing T cells, NK cells, intestinal intraepithelial lymphocytes, and lymphoid dendritic cells. CD8a functions primarily as a co-receptor for MHC class I molecule:peptide complex. CD8a is reported to interact simultaneously with the T-cell receptor (TCR) and the MHC class I proteins presented by antigen presenting cells. In NK-cells, the presence of CD8a homodimers at the cell surface provides a survival mechanism allowing conjugation and lysis of multiple target cells. CD8a homodimer molecules also promote the survival and differentiation of activated lymphocytes into memory CD8 T-cells. Cytoplasmic domains of CD8 provide binding sites for the tyrosine kinase lck, facilitating intracellular signaling events that lead to T cell activation, development, and cytotoxic effector functions. CD8+ cytotoxic T cells play an important role in inducing cell death of tumor cells, as well as cells infected by virus, bacteria or parasites. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
