General description

Purified Human IgG is isolated from pooled normal human serum by fractionation and ion-exchange chromatography. The product is supplied as an essentially salt-free (less than 1% sodium), lyophilized powder.

Application

IgG from human serum was used as a standard at a concentration of 1.274 mg/ml in ELISA to determine antibody responses to Porphyromonas gingivalis and Prevotella intermedia.[1] It was used for immunolabeling of Vero, BHK and PtK2 cells at a concentration of 0.2 mg/ml.[2] IgG from human serum was used as a standard in SDS-PAGE of anti-Shigella LPS antibodies.[3]

Biochem/physiol Actions

IgG antibody subtype is the most abundant of serum immunoglobulins of the immune system. It is secreted by B cells and is found in blood and extracellular fluids and provides protection from infections caused by bacteria, fungi and viruses. Maternal IgG is transferred to fetus through the placenta that is vital for immune defense of the neonate against infections.[4]

    1 answer
    1. The number of donors is not recorded for the original starting raw material. Historically, donor numbers for the plasma raw material are estimated to range from 1600 to 2000 individuals. No anti-inflammatory characterization has been performed. This product is intended for research use only. Unless otherwise stated this product is not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.

  3. What is the proportion of IgG1, IgG2, IgG3 and IgG4 in this product?

    1 answer
    1. In human serum, the 4 subclasses of human IgG are present at the approximate proportions of 66%, 23%, 7% and 4%, respectively.

      Helpful?

  4. How is this standard reconstituted?

    1 answer
    1. The product is essentially a salt-free preparation and should be reconstituted to a minimum concentration of 1 mg/ml with a saline solution containing 150 mM NaCl.

      Helpful?

  5. What is the expected MW of the product?

    1 answer
    1. The molecular weight of this product is not determined on a lot to lot basis. According to published literature, the molecular weight of Immunoglobulin G (IgG) is approximately 150 - 160 KDa. Historical data available for this product indicates that in SDS-PAGE electrophoresis this material will have multiple bands ranging from 155 kDa - 220 kDa.

  6. Necesito saber la secuencia de la proteína o el número para buscarla en uniprot

    1 answer
    1. The protein sequence is not determined for this product. Human IgG has 4 subclasses: IgG1, IgG2, IgG3 and IgG4. These are present in serum at approximately 66%, 23%, 7% and 4%, respectively.

      Helpful?

  7. What is the isoelectric point of this

    1 answer
    1. This product is not tested on a lot-to-lot basis to determine the isoelectric point. However, historical data indicates an isoelectric point range of 6.1 - 8.5.

      Helpful?

  8. Is product I4506, because it is derived from human serum, considered to potential biohazard?

    1 answer
    1. As indicated in the corresponding Data Sheet at the link below: "BIOHAZARD: Handle as if capable of transmitting infectious agents". The source material is tested and found negative for antibody to HIV, antibody to HCV, and for HbsAG.
