Gene Information

human ... GAB1(2549)

General description

Anti-GAB1 Antibody detects endogenous levels of total GAB1 protein.
The GRB2 associated binding protein 1 (GAB1) protein is encoded by the gene mapped to human chromosome 4q31.21. The encoded protein belongs to the Grb2-associated binding protein family. GAB1 protein structure contains pleckstrin homology domain and tyrosine residues. Anti-GAB1 Antibody detects endogenous levels of total GAB1 protein.

Immunogen

The antiserum was produced against synthesized peptide derived from human GAB1.

Immunogen Range: 644-674

Application

Anti-GAB1 antibody produced in rabbit has been used in proximity ligation assay (1:100) and western blotting.

Biochem/physiol Actions

GRB2 associated binding protein 1 (GAB1) acts as a scaffold protein for protein-protein interaction. It regulates growth factor receptors and enhances signal transduction of multiple intracellular signaling pathways, such as phosphatidylinositol 3-kinase and protein kinase B (PKB/AKT)/ mammalian target of rapamycin (mTOR) signaling pathways. Upregulated expression of GAB1 contributes to the progression of various types of cancer including human oral squamous cell carcinoma (OSCC), breast cancer, colorectal cancer, hepatocellular carcinoma, and ovarian cancer. Therefore, knockdown of the GAB1 gene can be considered as a potential therapeutic strategy for treating breast cancer and OSCC.

The Grb2-associated binding proteins are a family of platform proteins involved in signalling transduction pathways. A major family member, Gab1, modulates the effect of growth factor receptors and intracellular signalling pathways, including the phosphatidylinositol 3-kinase and AKT/mTOR signalling pathways. We

Despite extensive study of the EGF receptor (EGFR) signaling network, the immediate posttranslational changes that occur in response to growth factor stimulation remain poorly characterized; as a result, the biological mechanisms underlying signaling initiation remain obscured.
