General description

Immunoglobulin A (IgA) is the most abundant antibody isotype produced in humans. IgAs are heterogeneous and exist as two IgA subtypes. It has the ability to form multimers by interacting with the joining (J) chain.It is present in large quantities in blood. IgA antibodies protect mucosal surfaces against invading pathogens.
Specificity of the Alkaline Phosphatase Conjugated Anti-Human IgA is determined by Enzyme Linked Immunosorbent Assay (ELISA). Identity and purity of the antibody is established by immunoelectrophoresis (IEP) prior to conjugation. Electrophoresis of the antibody preparation followed by diffusion versus anti-goat IgG and anti-goat whole serum results in single arcs of precipitation.

Specificity

The conjugate is specific for human IgG. No cross-reaction with Mouse and Rat IgG.

Application

Anti-Human IgA (α-chain specific) Alkaline Phosphatase antibody produced in goat has been used in quantitative sandwich enzyme-linked immunosorbent assay (ELISA) and immunoassays.
Human milk samples were analyzed for the presence of IgA antibodies specific to certain antigens of H. pylori by western blot using alkaline-phosphate-conjugated goat anti-human IgA antibody.
Milk samples from 10 mothers were probed for bound milk IgA by western blot using alkaline-phosphatase goat anti-human IgA antibody.Elisa Assays were performed using alkaline phosphatase conjugated gaot anti-human IgA chain specific antibody (1:1000) to detect IgA immunoglobulins in serum and seminal plasma samples from infertile men.
The product may also be used for dot blot (1:30,000) and immunohistochemistry (at 1:100 dilutions, using formalin-fixed, paraffin-embedded sections).

Biochem/physiol Actions

IgA is a secretory glycoprotein antibody that regulates the physiological balance between commensal intestinal bacteria and the defenses of the host immune system. It is the highest generated antibody isotype. IgAs are mainly secreted in the intestinal mucosa and prevent the commensal microorganisms from invading the intestines
Immunoglobulin A (IgA) antibody is a secretory antibody present abundantly in mucous linings of gastrointestinal, respiratory and genitourinary tracts, tears and saliva. Thus, secretory IgA contributes to the humoral defense mechanism against the pathogens on mucosal surfaces.

Solution in 0.05 M Tris, pH 8.0, containing 1% bovine serum albumin, 1 mM MgCl2 and 15 mM sodium azide
