General description

PDGFs (platelet-derived growth factors) are disulfide-linked dimers consisting of two 12.0-13.5kDa polypeptide chains, designated PDGF-A and PDGF-B chains. Two distinct signaling receptors used by PDGFs have been identified and named PDGFR-α and PDGFR-β. PDGFR-α is high-affinity receptor for each of the three PDGF forms. On the other hand, PDGFR-β interacts with only PDGF-BB and PDGF-AB. The gene PDGF-B is mapped to human chromosome 22q13.1. Recombinant human PDGF-BB is a 24.3kDa disulfide-linked homodimer of two B chains (218 total amino acids).

Application

PDGF-BB human has been used in the cell viability assay. It has been used as a stimulator in PDGFRα (platelet-derived growth factor receptor α)-positive HTPCs (human testicular peritubular cells).

Biochem/physiol Actions

The three naturally occurring PDGFs (platelet-derived growth factors); PDGF-AA, PDGF-BB and PDGF-AB, are potent mitogens for a variety of cell types including smooth muscle cells, connective tissue cells, bone and cartilage cells, and some blood cells. The PDGFs are stored in platelet α-granules and are released upon platelet activation. The PDGFs are involved in a number of biological processes, including hyperplasia, chemotaxis, embryonic neuron development, and respiratory tubule epithelial cell development. PDGF-B along with PDGFR-β participates in vascular maturation. It brings smooth muscle cells and pericytes to growing vessels. Heterozygous mutation in PDGF-B is associated with primary familial brain calcification.

Lyophilized from 10 mM Acetic Acid.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a stabilizer (example 5% Trehalose) and store in working aliquots at -20°C to -80°C.
