General description

Monoclonal Anti-unmodified-Histone H3 (Lys9 ) (mouse IgG1 isotype) is derived from the hybridoma 9B1-2G6 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a peptide. Histone H3 is a core component of histone octamer of nucleosome along with other histones H4, H2A and H2B. Histone H3 comprises lysine and arginine residues in the N-terminal region. Conserved lysine residues in the N-terminal tail domains of histone H3 comprise theLys4, Lys9 and Lys27, which are the preferred sites of methylation.

Specificity

Monoclonal Anti-unmodified-Histone H3 (Lys9 ) recognizes human unmodified-Histone H3 (Lys9 ) (nontested in other species). This antibody also binds to peptides containing mono-, and di-methyl-Lys4 and acetyl-Lys14 of Histone H3. It does not bind to peptides containing mono-, di-, and tri-methyl-Lys9 and acetylLys9 of Histone H3.

Immunogen

Peptide corresponding to amino acids surrounding Lys9 of human Histone H3, conjugated to KLH.

Monoclonal Anti-unmodified-Histone H3 (Lys9) antibody produced in mouse has been used in:

Biochem/physiol Actions

Histones are subjected to extensive covalent modifications, including phosphorylation, methylation, acetylation and ubiquitination. These modifications are thought to play an important role in development and cancer. Histone H3 methylation at Lys9 is a modification intrinsically linked to epigenetic silencing and heterochromatin assembly. Histone methylation, as like acetylation, is a complex, dynamic process involving several processes, including transcriptional regulation, chromatin condensation, mitosis, and heterochromatin assembly. Moreover, lysine residues can be mono-, di-, and tri-methylated, adding further complexity to the regulation of chromatin structure.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8°C for up to one month. For extended storage,freeze at -20oC in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.
