General description

Vimentin is the major subunit protein of the intermediate filaments of mesenchymal cells. It is believed to be involved with the intracellular transport of proteins between the nucleus and plasma membrane. Vimentin has been implicated to be involved in the rate of steroid synthesis via its role as a storage network for steroidogenic cholesterol containing lipid droplets. Immunohistochemical staining for Vimentin is characteristic of sarcomas (of neural, muscle and fibroblast origin) compared to carcinomas which are generally negative. Melanomas, lymphomas and vascular tumors may all stain for Vimentin.

Specificity

Recognizes a 57-60 kDa protein. Shows no cross reactivity with other closely related intermediate filament proteins including desmin and GFAP. In tissue sections, this reagent shows positive reaction on connective tissue, endothelial cells, vascular smooth muscle cells and astrocytes. In tissue culture, typical intermediate filament profiles are seen in fibroblasts and other cell types, which contain vimentin

Purified vimentin from pig eye lens

Application

Anti-Vimentin Antibody, clone V9 is an antibody against Vimentin for use in IC, IH, IH(P) & WB.
Immunoblotting:
A dilution of 1:1000 was used in a previous lot.

Immunohistochemistry:
5-10 μg/mL was used on a previous lot; can be used on formalin fixed paraffin embedded tissues with traditional high temperature, citrate buffer treatments. Prolonged formalin fixation can reduce antivbody reactivity.

Immunocytochemistry:
5-10 μg/mL of a previous lot was used in IC.

Optimal working dilutions must be determined by end user.

Quality

Evaluated by Western Blot on HeLa lysates.

Western Blot:
1:1000 dilution of this antibody detected VIMENTIN on 10 μg of HeLa lysates
