General description

Expression of the POU-domain transcription factor Octamer-4 (Oct-4) is widely regarded as a hallmark of pluripotent stem cells. The relationship of Oct-4 to pluripotent stem cells is indicated by its tightly restricted expression to undifferentiated pluripotent stem cells. Upon differentiation to somatic lineages, the expression of Oct-4 disappears rapidly. Unlike the majority of pluripotent stem cell markers, the biological role of Oct-4 has been well characterized. Studies performed in mice point to the critical role of Oct-4 in the establishment and/or maintenance of pluripotent stem cells in an uncommitted state.

Specificity

Recognizes Oct4 (POU5F1). The molecular weight of the protein is 38,601 Daltons.

Synthetic peptide, amino acids 282-297of Oct4.

This Anti-Oct-4 Antibody is validated for use in IC, IH, WB for the detection of 41186.
Western blot: 1:200-1:1,000 using ECL. Oct-4 in cells typically shows ~40-44kDa band or doublet.

Immunohistochemistry: 1:200-1:1,000.

Immunocytochemistry: 1:200-1:1,000.

Optimal working dilutions must be determined by the end user.

Storage and Stability

Maintain unopened vial at -70°C for up to 6 months. Avoid repeated freeze/thaw cycles.

The rehydrated antibody solutions can be stored undiluted at 2–8°C for 2 months without any significant loss of activity. Note, the solution is not sterile, thus care should be taken if product is stored at 2–8°C. For storage at -20°C, the addition of an equal volume of glycerol can be used, however, it is recommended that ACS grade or higher glycerol be used, as significant loss of activity can occur if the glycerol used is not of high quality.

For freezing, it is recommended that the rehydrated antibody solution be further diluted 1:1 with a 2% BSA (fraction V, highest-grade available) solution made with the rehydration buffer. The resulting 1% BSA/antibody solution can be aliquoted and stored frozen at -70°C for up to 6 months. Avoid repeated freeze/thaw cycles.
