General description

Jagged-1 is a ligand for multiple Notch receptors and involved in the mediation of Notch signaling. It is involved in cell-fate decisions during hematopoiesis. Is involved in early and late stages of mammalian cardiovascular development. Inhibits myoblast differentiation. Enhances fibroblast growth factor-induced angiogenesis (in vitro). Defects in Jagged-1 are the cause of Alagille syndrome type 1 (ALGS1) and tetralogy of Fallot (TOF).
Jagged-1, encoded by the gene JAG1, is a Serrate homolog found in vertebrates. It is expressed by both primitive and mature human hematopoietic cells. The encoded protein contains EGF (epidermal growth factor) repeats, transmembrane domain and a cysteine-rich region in between the two.

Application

Jagged-1 has been used to activate Notch1 signaling pathway that modulates cardiac AMPK (5′ AMP-activated protein kinase) signaling cascade. Jagged-1 (human): FC (human) has been used to determine the cardiac substrate (fatty acid and glucose) metabolism in working heart model.

Biochem/physiol Actions

The encoded protein is a transmembrane-type ligand for Notch family receptor and is capable of inducing Notch signaling, which participates in embryogenesis, tissue regeneration, and carcinogenesis. Jagged-1 is a WNT-dependent Notch signaling activator that is involved in the maintenance of homeostasis of stem and progenitor cells. Jagged proteins are found to be involved in development and carcinogenesis. Overexpression of Jagged-1 has been observed in cervical and breast cancers.

Signal peptide and extracellular domain of human Jagged-1 (HJ1; CD339) (aa 1-1067) are fused at the C-terminus to the Fc portion of human IgG1.
