General description

Degradex® is made with poly(D,L-lactide-co-glycolide) with an L/G ratio of 50/50 and a Mw of 30,000.
To re-constitute:
  • Remove the product from freezer.
  • Wait until temperature rises to the ambient temperature.
  • Add DI water or buffer to the container.
  • Vortex and sonicate for 5 minutes to ensure the microspheres are well suspended.
The particle size of the PLGA nanoparticles are closely dependent on both the nature and concentration of the stabilizer and polymer.[1][2]

Application

Plain PLGA particles can be used for testing system compatibility in drug delivery research. Particles can be used as carriers by attaching drug or biological/chemical entities to particle surface. PLGA particles are used for controlled release, sustained release and targeted drug delivery.A study reports the use of PLGA nanoparticles as estrogen carriers. The carrier-drug system was prepared by emulsification-diffusion method.[1][2]

Questions

  1. Need more info on how to use the particles: How long are the PLGA nanoparticles stable for once reconstituted in water/PBS? Is 20mg/ml a good concentration to resuspend at? Presumably they should be stored at 4°C instead of refrozen once reconstituted?

    1. The stability of the PLGA nanoparticles in the solution is not determined. However, various sources state that suspensions may be stable for greater than 2 weeks in aqueous buffers when stored at 4°C. See the link below for preparation and surface conjugation instructions. Allow the product to reach room temperature before suspending it in water or buffer. Suspensions may require vertexing or sonication:
