Cell Line Description

TKE2 is the first cell line of mouse corneal epithelial progenitor cells isolated from the basal layer of the limbal epithelium.TKE2 has applications in the cell biology of corneal epithelial cells, toxicity, and differentiation. TKE2 can be induced to differentiate into stratified epithelium on increasing the calcium concentration and addition of foetal bovine serum.

Culture Medium

Stemline Keratinocyte Medium II (Sigma S0196) + Stemline Keratinocyte Growth Supplement (Sigma S9945) + 2 mM Glutamine.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:10, i.e., seeding at 2 x 102 - 2 x 104 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37 °C. It is important to inactivate the trypsin with an equal volume of trypsin inhibitor. To remove all traces of trypsin and trypsin inhibitor pellet the cells by centrifugation and resuspend the cells in fresh medium. Media change every second day.
