Specificity

Clone 1C10 is a ZooMAb® Rabbit recombinant monoclonal antibody that specifically detects Podocin. It targets an epitope within 19 amino acids from the C-terminal, cytoplasmic domain.

Immunogen

KLH-conjugated linear peptide corresponding to 19 amino acids from the C-terminal region of human Podocin.

Application

Quality Control Testing

Evaluated by Western Blotting in Human kidney tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Podocin in Human kidney tissue lysate.

Tested applications


Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected Podocin in human kidney tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Podocin peptide with a KD of 4.1 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Podocin in human kidney tissue sections.

Target description

Podocin (UniProt: Q9NP85; also known as NPHS2) is encoded by the NPHS2 gene (Gene ID: 7827) in human. Podocin is a member of the stomatin family, which consists of hairpin-like integral membrane proteins with intracellular NH2- and COOH-termini. It is exclusively expressed in the podocytes of fetal and mature kidney glomeruli. It is a raft associated component of the glomerular foot-process membrane where it is localized at the insertion of the slit-diaphragm in podocytes. It plays a role in the regulation of glomerular permeability, acting as a linker between the plasma membrane and the cytoskeleton. Two isoforms of Podocin have been described that are produced by alternative splicing. Isoform 1 is located in cell membrane and peripheral membranes and isoform 2 is present in endoplasmic reticulum. Knock-out mice that lack slit diaphragm die within the first weeks of life due to renal failure. Mutations in NPHS2 gene are linked to the development of a steroid resistant nephrotic syndrome (NPHS2) that is characterized by severe proteinuria, resulting in complications such as hypoalbuminemia, hyperlipidemia, edema, and end-stage renal failure in the first two decades of life. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
