Yes. When performing fluorescent staining procedures, aldehydes and alcohols are common fixatives. Paraformaldehyde solutions are prepared without the use of methanol. It is believed that methanol can adversely affect the staining of membrane bound proteins. It it for this reason that freshly prepared paraformaldehyde solutions are preferred over formaldehyde solutions that may contain up to 10 or 15% methanol to keep the formaldehyde from re-polymerizing back into solid paraformaldehyde. When using paraformaldehyde to fix tissue, frozen sections are preferred over paraffin processing.

Can I use paraformaldehyde to fix the tissue and then use fluorescence imaging? For example: Inject Tritc i.v. and then extract the brain tissue. The brain would be fixed in paraformaldehyde to be sliced later and imaged for fluorescence
