Description générale

Propidium iodide (PI) is often used for cell cycle analysis by flow cytometry. Compared to this analysis method, the BioTracker Far Red Live Cell Cycle Assay Solution does not require cell fixation or permeabilization, and is permeable in live cells. Use 5 uL of probe solution in 500 uL for each sample. Vial contains enough for 50 tests.IMPORTANT APPLICATION NOTES:*Wash cells thoroughly to remove trypsin. Residual trypsin can confound analysis of flow cytometry populations.*Adjust gates after cells are stained with probe SPECTRAL PROPERTIESExcitation: 405 - 407 nmEmission: 450/50 BPUse violet laser for excitation.

Description de la cible

The control mechanism of the cell cycle is closely linked to cell proliferation. The cell cycle includes cell division, in which one cell produces two daughter cells. The cell cycle can be assessed by detecting cells stained for DNA using flow cytometry, followed by analysis of the proportion of stained nuclei in cell cycle phase. The cell cycle is classified into two major phases: interphase and the mitotic (M) phase. Interphase is composed of G1 phase (preparation for DNA synthesis which occurs prior to cell division), S phase (DNA synthesis and replication), and G2 phase (chromosome duplication). M phase involves mitosis and cytokinesis. Cell cycle progression is regulated by cyclins and cyclin-dependent kinases. To determine the action of drugs such as anticancer therapeutics, it is essential to analyze the mechanism of cell-cycle checkpoint.

Unlike propidium iodide (PI), this probe is membrane permeable and highly specific for DNA, which makes it possible to analyze the cell cycle by simply adding the reagent to a cell suspension without any fixation, permeabilization, or other sample prep.
