Xanthine Oxidase (XO), a versatile molybdoflavoprotein, is one of the forms of xanthine oxidoreductases (XORs). XO is characterized with two flavin molecules (FAD), two molybdenum atoms, and eight iron atoms bound per enzymatic unit.[1]

Application

Xanthine Oxidase microbial has been used:
  • to determine in vitro xanthine oxidase activity[2]
  • as a source to generate superoxide anions during superoxide dismutase assay[3]
  • as a component of the reagent mix in colorimetric paper-based device to detect allopurinol in traditional medicine by enzymatic reaction

Biochem/physiol Actions

Also contains proteolytic activity.
Xanthine Oxidase catalyzes the hydroxylation of hypoxanthine to xanthine and xanthine to uric acid and oxygen free radicals during purine metabolism.[3][4] This enzyme is also implicated in caffeine metabolism.[5] XO also plays a vital role in the catabolism of xenobiotics including various drugs such as thiopurine drugs, containing 6-mercaptopurine, allopurinol, and uric acid.[6] Excess of XO activity causes hyperuricemia (higher levels of uric acid in the blood), which further leads to gout.[7]

Other Notes

One unit will convert 1.0 μmole of xanthine to uric acid per min at pH 7.5 at 25 °C. Approx. 50% of the activity is obtained with hypoxanthine as substrate.

  1. Good morning, I purchased xanthine oxidase X2252-25UN for quantifying SOD enzymatic activity. Which buffer should I use to solubilize the enzyme? After solubilization, how should I store the resulting solution?

    1. This product is soluble in water at a concentration of 10 mg/mL. It can also be dissolved in a buffer according to specific experimental references, such as 50 mM potassium phosphate. The stability of the solution has not been tested.

  2. Guten Tag, in welchem Lösungsmittel und bis zu welcher Konzentration ist das Enzym gut löslich? Kann Natrium Phosphat Puffer verwendet werden? Danke.

    1. For assay purposes, this product is prepared in 50 mM Potassium Phosphate Buffer, pH 7.5 at 25°C. The material is lyophilized from a solution containing BSA and sodium glutamate. While the solubility has not specifically been tested in sodium phosphate buffer, it is expected to be suitable. Please see the link below to review the assay protocol for this item:
