General description

El azul de tripano es un colorante utilizado para distinguir entre células vivas y células muertas. Es un colorante vital que no es absorbido por las células viables sanas, pero tiñe las células con una membrana celular dañada. De esta forma, sólo pueden contarse las células muertas. Este método suele denominarse método de exclusión de colorante.

El azul de tripano (TB), un tinte azoico hidrófilo aniónico, atraviesa solo las membranas celulares de las células muertas, tiñendo por tanto de azul las células y los tejidos muertos. Se utiliza en microscopia cuantitativa en una técnica de tinción de exclusión de colorante para diferenciar entre células de mamíferos vivas y muertas.[1]

Application

La disolución de azul de tripano se ha utilizado :
  • en el ensayo de viabilidad celular para contar células viables y muertas[2]
  • en el ensayo de rescate para contar células viables[2]
  • en el método de recuento por exclusión de azul tripano para determinar la curva de proliferación[3]
  • como control salino y en la preparación de la disolución de lipopolisacárido (LPS) para confirmar el éxito de la infusión de LPS[4]
  • en la prueba de exclusión de colorante azul tripano para determinar la viabilidad de las células epiteliales gástricas (GEC)[5]
  • para contar las células de GEC utilizando un hemocitómetro[5]

Preparation Note

Se prepara en cloruro de sodio al 0,81% y en fosfato de potasio dibásico al 0,06%.

    1. Trypan blue will stain cells that have disrupted membranes.  It cannot differentiate between permeabilized membranes caused by apoptosis or necrosis.

  4. Why is Trypan blue, Product T8154, used to determine cell viability?

    1. Trypan Blue is a vital stain recommended for use in estimating the proportion of viable cells in a population. The reactivity of this dye is based on the fact that the chromophore is negatively charged and does not react with the cell unless the membrane is damaged. Staining facilitates the visualization of cell morphology. Live (viable) cells do not take up the dye and dead (non-viable) cells do.

  5. Trypan blue, Product T8154, contains a precipitate.  Is it still good to use?

    1. The trypan blue solution is still good to use.  It can be warmed to dissolve the precipitate or it can be filtered to remove the precipitate.

  6. Why are there red fibers in Product T8154, Trypan blue solution?

    1. A report of fibers in Product T8154, Trypan blue solution is sufficiently common almost to be a characteristic of the product. These fibers are a cosmetic fault rather than one of functionality.  The presence of fibers may be more pronounced following cold storage or shipping.  This product should be stored at room temperature. Filtration to remove the fibers will not affect the use of the product.

  8. Do the cells need to be washed to remove the culture media prior to staining with Product T8154, Trypan Blue solution?

    1. Trypan Blue has a greater affinity for serum proteins than for cellular protein. If the background is too dark, cells should be pelleted and resuspended in protein-free medium or salt solution prior to counting.
