General description

Phosphatidylinositol-3-phosphate-5-kinase, type 3 (PIP5K3, also known as PIKfyve and FAB1) belongs to a large family of lipid kinases that modify the phosphorylation state of phosphatidylinositol (PtdIns). PIP5K3 has been shown to be localized on early and late endosomes.
Phosphatidylinositol-3-phosphate/phosphatidylinositol-5-kinase, type III (PIP5K3) is a constituent of a trimolecular protein complex. It is a 2089 amino acid protein. The gene encoding it is located on human chromosome 2 and has 41 exons.

Specificity

Anti-PIP5K3 (N-terminal) specifically recognizes human PIP5K3.

Anti-PIP5K3 (N-terminal) antibody produced in rabbit can be used in several immunochemical techniques including immunoblotting.

Biochem/physiol Actions

Phosphatidylinositol-3-phosphate-5-kinase, type 3 (PIP5K3) enzymatic activity is critical for cell morphology and endocytic membrane homeostasis. PIP5K3 acts as a negative regulator of endosome function or regulating endosome-trans-golgi-network (TGN) trafficking. It negatively regulates exocytosis on neurosecretory cells. PIP5K3 regulates the trafficking of IRAP/GLUT4 vesicles to plasma membrane in adipocytes, suggesting that it may play a pleiotropic role in membrane trafficking, including the exocytic pathway.
Phosphatidylinositol-3-phosphate/phosphatidylinositol-5-kinase, type III (PIP5K3) is involved in the synthesis of phosphatidylinositol 3, 5-bisphosphate. It also possesses a protein kinase activity. PIP5K3 regulates specific signaling pathways as well as membrane trafficking. Mutations in the gene encoding it are associated with fleck corneal dystrophy.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.
