General description

Fetuin from fetal bovine serum is a 64 kDa plasma glycoprotein. It is produced by the liver and adipose tissue.

Application

Fetuin is a protein with the ability to bind to the insulin receptor and inhibit insulin action. Fetuin has been used in studies of type 2 diabetes mellitus (T2DM). In patients with T2DM, fetuin has been shown to be significantly lower when compared to control patients. Fetuin has also been used in studies to demonstrate its protective role in the development of arterial calcification.
Fetuin from fetal bovine serum has been used:
  • to synthesize primary and secondary calciprotein particles (CPP)
  • to study the binding of Xerocomus chrysenteron lectin (XCL) to the T antigen expressed by membrane proteins
  • as a supplement in Ham′s F10 media for myoblast culture.

Biochem/physiol Actions

Fetuin, an inhibitor of insulin receptor tyrosine kinase, modulates bone remodeling and calcium metabolism. It is involved in keratinocytes migration and breast tumor cell proliferative signaling. Fetuin also regulates vascular calcification, insulin resistance and protease activity It acts as a biomarker for neurodegenerative diseases. Fetuin has anti-inflammatory property.

Preparation Note

Prepared by ammonium sulfate fractionation of fetal bovine serum by the method of Pederson, K.O., J. Phys. and Colloid Chem., 51, 164 (1947).

  1. Is there information available on the content/concentration of bovine IgG and BSA in the fetuin products F2379 and F3385 (CAS 9014-81-7)?

    1. Testing for bovine IgG and BSA is not conducted in the final product. A purity agarose gel is performed on F2379, but quantification, testing, or reporting for IgG and BSA is not done. Additionally, the marker size used for QC testing is uncertain, and both IgG and BSA are larger than fetuin.

  2. How pure is this fetuin? There's a protein content % but could there be other proteins from serum present when using ammonium sulfate fractionation?

    1. Product F2379 can not be considered pure Fetuin. Neither the purity, nor the identity of potential contaminating proteins, is determined.

  3. Is there a good antibody to recognize this bovine fetuin B by western blot?

    1. Unfortunately, the Fetuin antibodies currently available are not tested against bovine. While fetuin is fairly conserved from bovine to human, reactivity cannot be predicted. See the links below for fetuin antibodies against human that have been testing for use in western blot.
