General description

Platelet-derived growth factor (PDGF) is composed two polypeptide subunits of A and B chain. PDGF is produced by a number of cells including smooth muscle cells, endothelial cells and macrophages. The PDGF gene is highly conserved and is mapped to human chromosome 7p22.3. PDGF is structurally and functionally related to vascular endothelial growth factors.

Biochem/physiol Actions

Cell division and relocalization of pulmonary artery smooth muscle cells and fibroblasts is known to be triggered by platelet-derived growth factor (PDGF). Due to its proliferatory function, it is associated with fibroproliferative disorders such as atherosclerosis, pulmonary arterial hypertension and lung fibrosis. PDGF exists both in homo and heterodimeric forms and stimulates α and β-cell surface receptors. Vascular endothelial growth factor expression is regulated by PDGF-AA autocrine signalling and is known to promote tumor angiogenesis in vivo.
