Applicazioni

Quality Control Testing

Evaluated by Western Blotting in MDCK cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Collagen -4 in MDCK cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Collagen -4 in Human Kidney lysates.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Collagen -4 in human kidney tissue sections.

Affinity Binding Assay Analysis: The binding affinity of this antibody to Collagen -4 chain peptide was at least ten thousand-fold (10,000X) higher than that obtained with the non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Collagen -4 in HeLa cells.

Immunofluorescence Analysis: A 1:100 dilution from a representative lot detected Collagen -4 in human kidney tissue sections.

Collagen alpha-4(IV) chain (UniProt: P53420; also known as Collagen -4) is encoded by the gene COL4A4 (Gene ID: 1286) in human. Type IV collagen is the major structural component of glomerular basement membranes (GBM). Together with laminins, proteoglycans and entactin/nidogen it forms a network in the basement membrane, which is a specialized extracellular matrix structure that supports and separates different tissue compartments. COL4A4 is essential for maintaining the structural integrity and function of various tissues, particularly the basement membranes of the kidney glomerulus and inner ear. It contributes to the stability and filtration properties of the glomerular basement membrane, which is critical for normal kidney function. COL4A4 is predominantly expressed in tissues rich in basement membranes, such as the kidney, the cochlea, and the lung. Mutations in the COL4A4 gene disrupt the structure and function of the collagen IV network, leading to the deterioration of basement membranes in the kidneys and other affected organs. This ZooMAb^® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Stato fisico

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Stoccaggio e stabilità

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
