Descrizione generale

The gene KLHL20 (kelch like family member 20) is mapped to human chromosome 1q25.1. The encoded protein localizes to the trans-Golgi network (TGN). Small amount of the protein is also present in PML-NBs (promyelocytic leukemia - nuclear bodies). KLHL20 belongs to the BTB (broad-complex, tramtrack, and bric-à-brac) family of proteins.

Azioni biochim/fisiol

KLHL20 (kelch like family member 20) is a substrate adaptor of cullin3 (Cul3). It is required for post-Golgi trafficking. The Cul3-KLHL20 complex works as an ubiquitin E3 ligase and is responsible for polyubiquitination of coronin-7, thereby targeting coronin-7 to trans-Golgi network. KLHL20 is also involved in autophagy by regulating autophagy-associated degradation of ULK1 (unc-51 like autophagy activating kinase 1) and VPS34 (vacuolar protein sorting 34) complex subunits. Additionally, KLHL20 is associated with hypoxia responses and tumorigenesis. HIF1 (hypoxia inducible factor 1)-mediated upregulation of KLHL20 gene causes hypoxia-induced PML (promyelocytic leukemia) polyubiquitination. PML destruction is linked with epithelial-mesenchymal transition, tumor growth, angiogenesis and other related events.
