General description

Le laminine sono glicoproteine adesive non collagenose con un peso molecolare riguardevole. Si tratta di proteine eterotrimeriche con tre catene polipeptidiche legate mediante ponti disolfuro note come catene α, β eγ. I molti domini polipeptidici delle laminine possono legarsi a numerosi recettori superficiali della cellula, tra cui svariati dimeri di integrina, distroglicano e sindecani e altri componenti della matrice extracellulare (ECM). La laminina è una glicoproteina trimerica multidominio della matrice extracellulare ed è il principale componente delle lamine basali, strutture specializzate che fungono da impalcatura per l′adesione, la proliferazione e la differenziazione cellulare.

Application

La laminina della membrana basale estratta dal sarcoma di topo di Engelbreth-Holm-Swarm è utilizzata per:
  • differenziazione delle cellule produttrici di insulina (IPC)
  • saggi di inibizione dell'infezione
  • saggi di proliferazione
La laminina supporta la crescita e la differenziazione di molti tipi di cellule tra cui quelle epiteliali, endoteliali, neuronali, muscolari ed epatiche. Si consiglia l′uso come substrato per coltura cellulare alla dose di 1-2 μg/cm2. La concentrazione ottimale dipende dal tipo di cellula, nonché dall′applicazione e dagli obiettivi di ricerca.

Biochem/physiol Actions

Le lamininine sono una famiglia di glicoproteine che riveste una funzione integrante nell′organizzazione strutturale dei tessuti animali Si associano al collagene di tipo IV tramite l′ entactina e il perlecano e alle membrane cellulari attraverso recettori quali l'integrina, i complessi glicoproteici distroglicani e le glicoproteine del sistema dei gruppi sanguigni Lutheran. La laminina possiede domini attivi per il legame col collagene, l'adesione cellulare, il legame con l′eparina e con il frammento di crescita dei neuriti.

Preparation Note

Questo prodotto viene fornito ad una concentrazione di 1 − 2 mg/mL in Tris-HCl 50 mM, e NaCl 150 mM. E′ filtrato utilizzando un filtro da 0,2μm. Scongelare lentamente la soluzione a 2-8 °C prima dell'uso per evitare la formazione di gel. Per l'uso come rivestimento, diluire in una soluzione salina bilanciata, ricoprire la superficie di coltura con il volume minimo necessario e incubare a 37 °C per due ore. Lavare 3 volte con PBS prima di piastrare le cellule. Le superfici così rivestite di laminina possono essere conservate per un mese a 2-8 °C

Other Notes

La laminina è una glicoproteina trimerica multidominio della matrice extracellulare ed è il principale componente non collagenico della lamina basale che supporta l'adesione, la proliferazione e la differenziazione. La laminina è composta dalle catene A, B1 e B2, legate da molti legami disolfuro. Questa laminina è stata isolata dal tumore di topo di Engelbreth-Holm-Swarm.

  1. Can the plates to which I add laminin be stored for a certain amount of time—for example, a week in the refrigerator? The thing is, when I make them fresh, I sometimes miscalculate how many slices I need to use, and I always end up wasting some. Thankss

    1. Sterile laminin-coated plates/coverslips may be stored at 2–8°C for approximately 1 month. Store coated items sealed to avoid excess drying. Do not use if discoloration or spider-web formations are observed on the coated surface. Please see the link below to review additional information available in the product datasheet:

  2. How long can stocks be frozen at -20C for?

    1. In undiluted form or diluted to working concentration, this material may be stored at -20°C in aliquots up to the recommended retest date for the specific lot. This data is published in the Certificate of Analysis. Avoid repeated freeze/thaw cycles. The solution may be transferred to smaller vials in working volumes for long-term storage.     1. The product L2020 will be shipped using dry ice. Upon receipt, it should be stored at -20 degrees Celsius.

  4. How is this laminin concentration determined? (by which method?) I tried to measure the concentration of this product by ELISA, and the results always came out half of what is written on the vial.. what is the protein purity?

  5. Can I coat Laminin, Product L2020, on top of poly-L-lysine for better adherence?

    1. When coating coverslips or slides for use with neural stem cells (neurospheres), the following procedure is recommended. Precoat the coverslips with poly-L-lysine (Product No. P5899) at 100 μg/mL. This is incubated for 30 minutes or more at room temperature. The coverslips are then washed with sterile water and allowed to dry. The poly-lysine coated coverslips can be stored at room temperature until ready to use (up to ~1 month). Coverslips are then coated with 30 - 50 μL of warmed laminin, which is at a concentration of 50 μg/mL. Incubate at 37°C for 30 minutes. Remove the laminin, and wash with DME (50 μL) twice. Cells are then added to the coverslips.

  6. How can I store solutions of Laminin, Product L2020?

    1. The product has a recommended storage temperature of -20°C.  Please note, the product needs to be thawed slowly at 2-8°C.  If the product has been warmed up at room temperature and it gels, it cannot be reactivated for use. We recommend storage at 2-8°C for up to one week. For longer storage, the product needs to be frozen at -20°C in single-use aliquots.

  10. What type of laminin is Product L2020?

    1. In the nomenclature system by Burgeson, this is Laminin 1. The laminins were numbered with Arabic numerals in the order discovered. The previous A, B1, and B2 chains, and their isoforms are listed by the Greek letters, for alpha, beta, and gamma, respectively, followed by an Arabic numeral to identify the isoforms.  For example, the first laminin identified from the Engelbreth-Holm-Swarm tumor (EHS) was designated laminin-1 with the chain composition α-1/β-1/γ-1. The genes for these 3 chains are LAMA1, LAMB1, and LAMC1. Reference: Burgeson, R.E., et al., A new nomenclature for the laminins. Matrix Biol., 14, 209-211 (1994).

    work very well

    Both 2°C~8°C overnight coating and rapid coating(37C, 2hrs.) methods work perfectly fine
