Gene Information

General description

Fibrinogen was the first blood coagulation factor to be identified, and it is a precursor of fibrin protein. This protein is composed of two chains, each of which is composed of three chains- α, β and γ. These three subunits are coded by three genes located in the human chromosomal locus 4q23-q32, and spans 51kb.[1]

Application

Fibrinogen from human plasma has been used for-
  • fibrinogen adsorption assay performed on swCNTs (single-walled carbon nanotubes)
  • the characterization of fibrinogen structure in the presence of CNT using CD (Circular dichroism) spectroscopy and temperature scan[2]
  • the preparation of RBC suspension obtained from venous blood of antecubital vein, to assess RBC aggregability[3]
  • fibrinolysis assay performed on mouse embryonic fibroblasts (MEFs)[4]
  • the preparation of fibrinogen-labeled gold (Fgn/Au)[5]
  • the quantification of in vitro angiogenesis by human microvascular endothelial cells[6] and
  • the assessment of fibrin-induced apoptosis and differentiation-regulation in trophoblast obtained from term human placentas.[7]

Biochem/physiol Actions

Il fibrinogeno è una proteina della fase acuta che fa parte delle proteine della cascata della coagulazione. Il risultato finale della cascata è la produzione di trombina che converte il fibrinogeno in fibrina. La trombina proteolizza rapidamente il fibrinogeno con rilascio del fibrinopeptide A. La perdita di questo piccolo peptide non è sufficiente a rendere insolubile la molecola di fibrina risultante ma tende a formare complessi con molecole di fibrina e fibrinogeno adiacenti. La trombina poi scinde un secondo peptide, il fibrinopeptide B, dalla fibrina e i monomeri di fibrina creati polimerizzano in seguito spontaneamente formando un gel insolubile. La fibrina polimerizzata è tenuta insieme da forze non covalenti ed elettrostatiche ed è stabilizzata dall′enzima transamidante, fattore XIIIa, che è prodotto dall′azione della trombina sul fattore XIII. Gli aggregati insolubili di trombina (coaguli) e le piastrine aggregate bloccano poi il vaso danneggiato e impediscono l′ulteriore sanguinamento. La quantità di fibrinogeno nel plasma può servire da indicatore aspecifico della eventuale presenza di un processo infiammatorio nell′organismo. Il fibrinogeno originato da qualsiasi mammifero sarà scisso dalla trombina di qualsiasi mammifero.

Fibrinogen is an acute-phase inflammatory protein, which plays an essential role in blood clotting. Coronary heart disease is linked to the plasma levels of fibrinogen in Chinese population, and might have potential as a marker and risk factor for the same.[1] A -455 allele of fibrinogen β (FGB) chain is linked with poor survival in Caucasian women with Finnish stroke.[8] Ischemic stroke preceded by hyperfibrinogenemia has poor outcome, which is independent of the baseline fibrinogen concentration.[9]

    1. This product has not been tested for freeze-thaw stability. However, in powder form, repeatedly removing this product from a freezer for the purpose of preparing fresh solutions, is not expected to be detrimental to the protein. Prepared solutions may be stored up to 1 week at 2 - 8 °C. It is not recommended to freeze stock solutions, as the freeze-thaw process may lead to protein degradation.

  2. F4883-500MG contains Sodium Citrate and Sodium Chlorure. Is it possible to have their concentration ? What is the volume of sodium chloride we have to put in the bottle please?

    1. This material is lyophilized from a solution of 75 mM NaCl (4.39 mg/mL), 10 mM Na Citrate (2.94 mg/mL). It can be solubilized in 0.85% (w/v) sodium chloride to 2 mg/ml, by layering the protein on top of the saline solution and placing it in a 37°C water bath for 4-6 hours, with gentle agitation.

  3. Hello. For your F4883-500MG Fibrinogen product, is there 500mg of pure fibrinogen in the vial? Or is there 500 mg of reagent - of which a fraction is clottable Fibrinogen? Do you have preparation instructions for this reagent?

    1. F4883-500MG contains 500 mg of solid. Fibrinogen can be solubilized in 0.85% (w/v) sodium chloride (2 mg/ml) by layering the protein on top of the saline solution and placing it in a 37 °C water bath for 4-6 hours with gentle agitation.
