General description

La 2′,7′-diclorodiidrofluoresceina diacetato (DCFH-DA) è un colorante fluorescente sensibile all'ossidazione, utilizzato per rilevare il livello intracellulare di specie reattive dell'ossigeno (ROS). Il DCFH-DA è una sonda non fluorescente in grado di permeare le cellule, che può essere idrolizzata dall'esterasi intracellulare a dare DCFH, che per ossidazione da parte dei ROS intracellulari forma DCF fluorescente.
La lunghezza d'onda di eccitazione di questo colorante è 504 nm, mentre quella di emissione è 529 nm. L'impiego del DMSO come solvente non è stato confermato ufficialmente, ma diverse pubblicazioni scientifiche suggeriscono che potrebbe essere adatto.

Application

  1. Is this product compatible with EGFP transfected cells?

    1. This product (DCFH-DA) is cell-permeable and converts intracellularly to the fluorescent DCF upon ROS oxidation, enabling ROS detection in EGFP-transfected cells. However, direct fluorescence in the GFP channel is not ideal due to spectral overlap between DCF (Ex ~504 nm, Em ~529 nm) and EGFP (Ex ~488 nm, Em ~507–509 nm), which can interfere with signal and ROS quantification. If EGFP-transfected cells are used, include controls such as EGFP-only cells, DCFH-DA-only cells, unstained cells, and a positive ROS control. Alternatively, consider a red/far-red ROS probe to avoid spectral overlap.
