Description générale

NLRC4 (NLR family, CARD domain containing 4) is a cytoplasmic immune receptor protein associated with inflammatory responses. It is a member of the nucleotide-binding site and leucine-rich repeat (NBS-LRR) protein. It consists of ATP/GTPase-specific P-loop and Mg2+-binding site motifs. The NBs domain contains a nucleotide-binding pocket for ATP/dATP.

Application

The Human Protein Atlas project can be subdivided into three efforts: Human Tissue Atlas, Cancer Atlas, and Human Cell Atlas. The antibodies that have been generated in support of the Tissue and Cancer Atlas projects have been tested by immunohistochemistry against hundreds of normal and disease tissues and through the recent efforts of the Human Cell Atlas project, many have been characterized by immunofluorescence to map the human proteome not only at the tissue level but now at the subcellular level. Actions biochimiques/physiologiques

NLRC4 (NLR family, CARD domain containing 4) is involved in the inflammatory responses. Activated NLR (NOD-like receptors) transports the caspase-1 within the inflammasome for its proteolytic activation. It converts the inactivated pro-IL-1 into active caspase by autoproteolysis. This multiprotein caspase-1 generates several innate immune sensors such as interleukin-1 family cytokines (IL1FCs), followed by cytokine productions and inflammatory cell death (pyroptosis). Mutation in the NLRC4 causes an auto-inflammatory syndrome of enterocolitis and autoinflammation.
