Specificity

Clone 2G10 is a ZooMAb rabbit recombinant monoclonal antibody that specifically detects human Urokinase plasminogen activator surface receptor (uPAR).

Immunogen

A recombinant fragment corresponding the first 277 amino acids from human Urokinase plasminogen activator surface receptor (uPAR).

Application

Anti-uPAR, clone 2G10 ZooMAb, Cat. No. ZRB1224, is a Rabbit monoclonal antibody that targets uPAR and has been tested for use in Flow Cytometry, Immunocytochemistry, Immunohistochemistry, Inhibition Assay, and Western Blotting.
Inhibits Activity/Function: A representative lot of this antibody inhibited the invasion of MDA-MB-231 cells (Harel, E., et al., 2014 UCSF Scientific Poster).

Flow Cytometry Analysis: 0.1 μg from a representative lot detected uPAR in one million SH-SY5Y cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected uPAR in human lung cancer tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected uPAR in HT1080 cells.

Note: Actual optimal working dilutions must be determined by the end user as specimens and experimental conditions may vary.

Target description

Urokinase plasminogen activator surface receptor (UniProt: Q03405; also known as U-PAR, uPAR. Monocyte activation antigen Mo3, CD87) is encoded by the PLAUR (also known as MO3, UPAR) gene (Gene ID: 5329) in human. uPAR is a GPI-anchored cell membrane receptor for urokinase plasminogen activator (uPA), an enzyme responsible for the activation of plasmin and degradation of extracellular matrix components. It is also a subject to negative-feedback regulation by U-PA, which cleaves it into an inactive form. uPAR is synthesized with a signal peptide (aa 1-22) and a propeptide (aa 306-335), which are subsequently cleaved off to produce the mature form. The mature form has three homologous domains known as DI, DII, and DIII. Higher expression of uPAR has been reported in many human cancers, including non-small cell lung cancer and colorectal cancer and it correlates with a poor prognosis and early invasion and metastasis. uPAR, through integrins interaction, is reported to play a crucial role in activating Src, FAK and paxillin, which are considered as critical mediators of cell migration. In addition, via its interaction with chemotaxis receptors for fMet-lleu-Phe, it mediates both uPA and fMLF-dependent cell migration. uPAR has also been implicated in cross-talk with EGFR, which mediates the shift from tumor cell dormancy to proliferation. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
