Specificity

Clone 1C9 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects beta-Tubulin. It targets an epitope within 22 amino acids from the C-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 22 amino acids from the C-terminal half of human β-Tubulin.

Application

Quality Control Testing

Evaluated by Western Blotting in Raji cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected β-Tubulin in Raji cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected β-Tubulin in Jurkat cell lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected β-Tubulin in human tonsil tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound β-Tubulin peptide with a KD of 2.5 x 10-8 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected β-Tubulin in A431 and HepG2 cells.

Target description

Tubulin beta-1 chain (UniProt: Q9H4B7) is encoded by the TUBB1 gene (Gene ID: 81027) in human. Tubulin is one of several members of a small family of globular proteins, which polymerize to form microtubules. The most common members of the tubulin family are alpha-tubulin and beta-tubulin that assemble onto microtubules forming large polymers. During polymerization the /β-tubulin heterodimers arrange into linear protofilaments that associate laterally to form the hollow microtubule cylinders. Within a protofilament, the tubulin heterodimers associate in a head-to-tail fashion, which makes microtubules intrinsically polar. The -tubulin within the dimer is oriented toward the plus end and the β-tubulin subunit toward the minus end. Tubulin binds two moles of GTP, one at an exchangeable site on the beta chain and one at a non-exchangeable site on the alpha chain. GTP-binding site of beta-tubulin is localized to amino acids 140-146. Multiple isoforms of β Tubulin have been described in mammalian tissues. β1 and β4 are reported to be present throughout the cytosol, β2 is localized in the nuclei and nucleoplasm, and β3 is a neuron-specific cytoskeletal protein. β1-tubulin is the most divergent at the amino acid sequence level. It is expressed exclusively in megakaryocytes and platelets in humans and plays an important role in the formation of platelets. It appears late in differentiation and localizes to microtubule shafts and coils within proplatelets. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitution

300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
