Biochem/physiol Actions

Erythropoietin (EPO) is a glycoprotein hormone which is essential for the proliferation of erythropoietic progenitors to produce red blood cells (RBCs). Peritubular fibroblast in kidney cortex is the primary site for EPO production in adults, though spleen, bone marrow and liver contribute to the production. In fetus, liver is the primary site for EPO production. Additionally, EPO functions in some cytoprotective activities like, antiapoptotic, antioxidant, and anti-inflammatory responses. Hypoxia inducible factors upregulate EPO expression. EPO expression increases with decreased hemoglobin concentration in few cases of anemia. Decreased production of EPO is the cause of anemia in chronic kidney diseases. Circulating endogenous antibodies to EPO (anti-EPO) is the cause of anemia in multiple diseases like human immunodeficiency virus (HIV) infection, hepatitis C virus infection, systemic lupus erythematosus. Gain-of-function polymorphism in EPO causes familial erythrocytosis.
