General description

Phosphorylase b is a dimer, usually exists in inactive form in the skeletal muscles.[1][2] Equilibrium exists between an active relaxed (R) state and a less active tense (T). Phosphorylase b favors the T state.[3] The enzyme possesses three domains, the N-terminal domain, glycogen-binding domain and the C-terminal domain.[1]

Application

Phosphorylase b from rabbit muscle has been used as a molecular weight marker in 12% polyacrylamide gel for keratinase,[4] and stress-70 protein.[5]
Phosphorylase b from rabbit muscle is to be used as a marker in SDS-PAGE. Phosphorylase b is used during chemical cross-linking studies as a SDS-PAGE molecular weight standard[6][7].

The amino acid composition is:Asp5l, Asn45, Thr35, Ser29, Glu64, Gln31, Pro36, Gly48, Ala63, Cys9, Val62, Met2l, Ile49, Leu79, Tyr36, Phe38, Trp12, Lys48, HiS22, and Arg63. Reference for the sequence and amino acid composition: K. Titani et al., Proc. Nat. Acad.
